چکیده

A simple framework for the analysis of the effects of errors in the method of isomorphous replacement is developed. This framework is used to derive phase probability distributions similar to those given by Blow & Crick [Acta Cryst. present analysis suggests an improved means of calculating the mean-square 'lack-of-closure' residuals and shows that they differ by a factor of two for centric and acentric reflections. It is also shown that the lack-of-closure residuals have a straightforward interpretation and that they may be used to estimate the error in the heavy-atom model and the degree of isomorphism between native and derivative structures if anomalous differences have been measured.

Application of the complex multivariate normal distribution to crystallographic methods with insights into multiple isomorphous replacement phasing.

Probabilistic methods involving maximum-likelihood parameter estimation have become a powerful tool in computational crystallography. At the centre of these methods are the relevant probability distributions. Here, equations are developed based on the complex multivariate normal distribution that generalize the distributions currently used in maximum-likelihood model and heavy-atom refinement. ...

Correlated phasing of multiple isomorphous replacement data.

Substantial highly correlated differences sometimes exist between a series of heavy-atom derivatives of a macromolecule and the native structure. Use of such a series of derivatives for phase determination by multiple isomorphous replacement (MIR) has been difficult because MIR analysis has treated errors as independent. A simple Bayesian approach has been used to derive probability distributio...

Enantiomorphous phase doublets from isomorphous replacement or anomalous scattering

A direct-methods procedure has been proposed for separating the phase doublet resulting from the use of either isomorphous replacement or anomalous scattering techniques. The phase doublet is expressed as ~#u= ~h±lA~.l. Formulae combining the structure-factor relationships with the phase-doublet information are given. Problems concerning the practical applications are also discussed. A test cal...

On integrating the techniques of direct methods and SIRAS: the probabilistic theory of doublets and its applications.

The mathematical formalism of direct methods is here applied to the SIRAS (single-isomorphous replacement combined with anomalous scattering) case. Specifically, the joint probability distribution of three structure factors, which plays the central role in the probabilistic theory of the two-phase structure invariants (doublets), is derived. This distribution leads directly to the conditional p...

Modeling Leukemia in Children Using Phase-type Distribution

Background: In this study, with the aim of modeling Leukemia in children using Phase-type distribution, three transitional phases including diagnosis, brain metastasis and testis/ovary metastasis, and one absorotion phase of recovery/death have been considered. The distribution was fitted and the probabilities of death or recovery were determined based on the independent variab...
